چکیده
Some studies demonstrate that gallic acid (GA) and myricetin (MYR) isolated from Rhus trilobata provide the therapeutic activity of this plant against cancer. However, few reports both compounds could also have potential in ovarian Therefore, evaluating cytotoxic GA MYR cancer cells determining possible action mechanism present are important. For purpose, SKOV-3 (ovarian adenocarcinoma; HTB-77™, ATCC®) were cultivated according to supplier’s instructions (37 °C 5% CO2) determine biological by confocal/transmission electron microscopy, PI-flow cytometry, H2DCF-DA, MTT, Annexin-V assays. Possible molecular targets determined Similarity Ensemble approach. Results showed treatments decreased viability at 50 166 μg/mL, respectively (p ≤ 0.05, ANOVA vs. vehicle group). They induced morphological changes (cytoplasmic reduction, nuclear chromatin condensation, cytoplasmic vesicles increment, polymerized actin, stabilized tubulin), cell cycle arrest (GA: 8.3% G2/M MYR: 78% G1), apoptosis induction 18.9% 8.1%), due ROS generation (34 42%) for 24 h In silico demonstrated interact with carbonic anhydrase-IX PI3K, respectively. conclusion, show through production, which modifies cytoskeleton induces apoptosis. be considered as base development new chemotherapy

Cytotoxic activity of caffeic acid and gallic acid against MCF-7 human breast cancer cells: An in silico and in vitro study
Objective: Phenolic compounds have been considered inhibitors of various cancers. Material and Methods: In this study, caffeic acid and gallic acid were appraised for their possible effects on apoptotic genes expression in a breast cancer cell line in vitro. We also evaluated ligand interaction and ligand binding with estrogen receptor alpha by m...
متن کاملProduction of Reactive Oxygen Species by Mitochondria
The mitochondrial respiratory chain is a major source of reactive oxygen species (ROS) under pathological conditions including myocardial ischemia and reperfusion. Limitation of electron transport by the inhibitor rotenone immediately before ischemia decreases the production of ROS in cardiac myocytes and reduces damage to mitochondria. We asked if ROS generation by intact mitochondria during t...
متن کاملReactive oxygen species mediate lysophosphatidic acid induced signaling in ovarian cancer cells.
Lysophosphatidic acid (LPA) is produced by tumor cells and is present in the ascites fluid of ovarian cancer patients. To determine the role of endogenous LPA in the ovarian cancer cell line SKOV3, we treated cells with the LPA receptor antagonist VPC32183 and found that it inhibited cell growth and induced apoptosis. Exogenous LPA further stimulated ERK and Akt phosphorylation and NF-κB activi...
متن کاملPGC1α induced by reactive oxygen species contributes to chemoresistance of ovarian cancer cells
Malignant cells are subjected to high levels of oxidative stress that arise from the increased production of reactive oxygen species (ROS) due to their altered metabolism. They activate antioxidant mechanisms to relieve the oxidative stress, and thereby acquire resistance to chemotherapeutic agents. In the present study, we found that PGC1α, a key molecule that both increases mitochondrial biog...
